The journey from Kuala Lumpur to Malacca takes about 2 to 2.5 hours, and from Malacca to Johor Bahru immigration takes another 2.5 to 3 hours before heading to Singapore.

You will remain in the vehicle during the immigration checkpoint clearance as the driver will use the car lane to enter Singapore. You may need to hand over your white card to the officer at the counter, and the driver will guide you through the process.
